I'm going to purchase an epiphone sheraton II, a left handed one.

I have played a righty one, and I really liked the feel and sound of it.

I'm just worried that it will be very muddy for what i listen to. (Pixies, R.E.M., White Stripes)

I have always liked Peter Buck's Rickenbacker tone, but thats a different story.

Is there any way to get like a single coil pickup on there? To get a brighter sound?

One chord is fine.
Two you're pushing it.
Three and you're into jazz.

You can put some humbucker-size P90s in there (maybe too growly), or get some of the Gretsch-style humbuckers which are much brighter and not as warm as what you'll hear with Gibson / Epi humbuckers.

The GFS Memphis pickups (Gretsch knockoffs) are supposed to have a jangly Ric-like sound, especially in a semi-hollow guitar body, and are relatively inexpensive:

Also does it matter if you put a regular pickup in a lefty guitar?

Never even heard of a lefty pup. A "righty" will work.

That's a great idea for marketing, lefty pickups. Identical to righty pickups, except for the label. Just like Rogaine for Women, that comes in a pink box.

Need some lefty strings to go with those, sir?

They have a left handed pickup for Strat's because of the 'stagger' on certain models.
(See picture for example of staggered pole pieces)

They are used to get the correct stagger placement on left handed guitars, or to get the 'Jimi Hendrix' stagger placement
on right handed guitars.

The wires are just on the wrong side more or less.

So basically....
