I have a Fender Custom Shop '60 Closet Classic.
Would my guitar sound improve if I change the pickups?
I've heard Lindy Fralin Vintage Hots or John Suhr's
V60LP are good. Will they make a big enough difference
in sound to justify the expense?

Hard to believe that a Fender Custom Shop '60 Closet Classic would come with crap pickups. Maybe it's just not the sound that you are looking for.
Changing pickups will certainly change the sound, but it's a matter of opinion as to whether or not it would improve your guitar sound. You may want to take a look at EMG pickups.
