how to tell what is the 'root'

I've come up with a little lick, and would like to know how I can build chords on those notes, and what would the root be? The notes and sequence are E F# G A B C B A G F# E B

The notes that you have there fit the scale of D Mixolydian, which is a mode of G major. D mixolydian is a major scale with a flat vii.

To harmonise all the notes in the mixolydian scale, you would have Dmaj, Emin, Fdim5, Gmaj, Amin, Bmin, Cmaj (I hope I haven't made any mistakes, there).

I started with nothing - and I've still got most of it left.

Since the only sharp note you have there is the F#, then those notes would correlate to the Key of G. The only note missing in your lick that you would find in the Key of G is the D note. The 3 primary chords in the Key of G would be G, C & D. The 3 secondary chords would be Am, Bm & Em. The dominate 7th would be D7.
